Italian term

a pennello

Italian to English Other Art, Arts & Crafts, Painting
Iscrizioni sacra, Latino; a pennello – lettere capitali

This is found in the description of a mural painting.

Can I say "by brush/paintbrush"? I also found "perfectly" as the translation for "a pennello."

with a paintbrush

I think it means "letters painted with a paint brush", or even "letters painted by brush" to a avoid the repitition of paint.

In your case, it's simply saying how the inscription is applied.
